During the last three years, more than 300 landraces belonging to different plant species have been the main focus of an Italian valorization research project (AgroBiodiversità Campana, ABC) aiming at analyzing, recovering, preserving, and collecting local biodiversity. In this context, phytosanitary investigation plays a key role in identifying potential threats to the preservation of healthy seeds in gene banks and the successful cultivation of landraces. The surveillance carried out in this study, in addition to highlighting the expected presence of common species-specific pathogens such as Ascochyta pisi in peas, Ascochyta fabae in broad beans, and Macrophomina phaseolina, Xanthomonas axonopodis pv. phaseoli, and Xanthomonas fuscans subsp. fuscans in beans, pointed to the presence of novel microorganisms never detected before in the seeds of some hosts (Apiospora arundinis in common beans or Sclerotinia sclerotiorum and Stemphylium vesicarium in broad beans). These novel seedborne pathogens were fully characterized by (i) studying their morphology, (ii) identifying them by molecular methods, and (iii) studying their impact on adult crop plants. For the first time, this study provides key information about three novel seedborne pathogens that can be used to correctly diagnose their presence in seed lots, helping prevent the outbreaks of new diseases in the field.

The diagnostic survey of seedborne fungal pathogens is fundamental for symptomless material stored in gene banks to avoid the diffusion of pathogens by germplasm distribution and propagation. In this work, seeds of Southern Italian landraces of the common bean (Phaseolus vulgaris L.) belonging to the gene bank at CREA (Italy) were inspected to assess their phytosanitary status. The phytopathological analysis revealed the presence of the most common pathogens associated with common bean seeds such as Fusarium spp., Macrophomina phaseolina, Rhizoctonia solani, Colletotrichum lindemuthianum and Diaporthe/Phomopsis complex. However, new fungi able to completely inhibit seed germination were also observed. The most aggressive were isolated, and the morpho-pathological characterization, DNA sequencing and phylogenetic analysis allowed us to define the strains as Botryosphaeria dothidea CREA OF 360.4 and Diplodia mutila CREA OF 420.36. These two plant pathogens are generally associated with grapevines and other fruit trees. Pathogenicity tests were carried out along with a transmissibility test in which the transmission of the pathogens to the seedlings was proven. Host range experiments revealed the ability of these pathogens to infect crops such as pepper and melon. To our knowledge, this is the first time that B. dothidea and D. mutila were detected on the common bean.

Sustainable agricultural systems based on the application of phyto-friendly bacteria and fungi are increasingly needed to preserve soil fertility and microbial biodiversity, as well as to reduce the use of chemical fertilizers and pesticides. Although there is considerable attention on the potential applications of microbial consortia as biofertilizers and biocontrol agents for crop management, knowledge on the molecular responses modulated in host plants because of these beneficial associations is still incomplete. This review provides an up-to-date overview of the different mechanisms of action triggered by plant-growth-promoting microorganisms (PGPMs) to promote host-plant growth and improve its defense system. In addition, we combined available gene-expression profiling data from tomato roots sampled in the early stages of interaction with Pseudomonas or Trichoderma strains to develop an integrated model that describes the common processes activated by both PGPMs and highlights the host's different responses to the two microorganisms. All the information gathered will help define new strategies for the selection of crop varieties with a better ability to benefit from the elicitation of microbial inoculants.

Climate change has led to the spread of plant pathogens in novel environments, causing dramatic crop losses and economic damage. Botryosphaeriaceae represents a massive fungal family, containing a huge number of plant pathogens, which are able to infect several hosts. Among them, Macrophomina phaseolina is a necrotrophic fungus, responsible for several plant diseases, including the soft stem rot of common bean, crown rot on strawberry and charcoal rot of several legumes. Here, Macrophomina, causing crown charcoal rot in chickpeas, was isolated from symptomatic plants in Cicerale (SA), Campania, South Italy. Morphological and molecular characterization was carried out and pathogenicity tests were performed. Phylogenetic analyses were performed comparing Macrophomina strains coming from different geographic areas and hosts. The experiments confirmed the pathogenicity of the isolate CREA OF 189.2 on chickpea, while host range highlighted the polyphagous nature of this strain; thus, symptoms were reported on lentils, common bean and cantaloupe. The multidisciplinary approach allows us to increase the knowledge about this emerging pathogen. To the best of our knowledge, this is the first report on Macrophomina phaseolina from chickpeas in Italy.

Food legumes are crucial for all agriculture-related societal challenges, including climate change mitigation, agrobiodiversity conservation, sustainable agriculture, food security and human health. The transition to plant-based diets, largely based on food legumes, could present major opportunities for adaptation and mitigation, generating significant co-benefits for human health. The characterization, maintenance and exploitation of food-legume genetic resources, to date largely unexploited, form the core development of both sustainable agriculture and a healthy food system. INCREASE will implement, on chickpea (Cicer arietinum), common bean (Phaseolus vulgaris), lentil (Lens culinaris) and lupin (Lupinus albus and L. mutabilis), a new approach to conserve, manage and characterize genetic resources. Intelligent Collections, consisting of nested core collections composed of single-seed descent-purified accessions (i.e., inbred lines), will be developed, exploiting germplasm available both from genebanks and on-farm and subjected to different levels of genotypic and phenotypic characterization. Phenotyping and gene discovery activities will meet, via a participatory approach, the needs of various actors, including breeders, scientists, farmers and agri-food and non-food industries, exploiting also the power of massive metabolomics and transcriptomics and of artificial intelligence and smart tools. Moreover, INCREASE will test, with a citizen science experiment, an innovative system of conservation and use of genetic resources based on a decentralized approach for data management and dynamic conservation. By promoting the use of food legumes, improving their quality, adaptation and yield and boosting the competitiveness of the agriculture and food sector, the INCREASE strategy will have a major impact on economy and society and represents a case study of integrative and participatory approaches towards conservation and exploitation of crop genetic resources.

Research has been increasingly focusing on the selection of novel and effective biological control agents (BCAs) against soil-borne plant pathogens. The large-scale application of BCAs requires fast and robust screening methods for the evaluation of the efficacy of high numbers of candidates. In this context, the digital technologies can be applied not only for early disease detection but also for rapid performance analyses of BCAs. The present study investigates the ability of different Trichoderma spp. to contain the development of main baby-leaf vegetable pathogens and applies functional plant imaging to select the best performing antagonists against multiple pathosystems. Specifically, sixteen different Trichoderma spp. strains were characterized both in vivo and in vitro for their ability to contain R. solani, S. sclerotiorum and S. rolfsii development. All Trichoderma spp. showed, in vitro significant radial growth inhibition of the target phytopathogens. Furthermore, biocontrol trials were performed on wild rocket, green and red baby lettuces infected, respectively, with R. solani, S. sclerotiorum and S. rolfsii. The plant status was monitored by using hyperspectral imaging. Two strains, Tl35 and Ta56, belonging to T. longibrachiatum and T. atroviride species, significantly reduced disease incidence and severity (DI and DSI) in the three pathosystems. Vegetation indices, calculated on the hyperspectral data extracted from the images of plant-Trichoderma-pathogen interaction, proved to be suitable to refer about the plant health status. Four of them (OSAVI, SAVI, TSAVI and TVI) were found informative for all the pathosystems analyzed, resulting closely correlated to DSI according to significant changes in the spectral signatures among health, infected and bio-protected plants. Findings clearly indicate the possibility to promote sustainable disease management of crops by applying digital plant imaging as large-scale screening method of BCAs' effectiveness and precision biological control support.

Plants produce a huge array of secondary metabolites that play a key role in defense mechanisms against detrimental microorganisms and herbivores, and represent a suitable alternative to synthetic fungicides in sustainable agriculture. In this work, twelve crude hydroethanolic extracts derived from leaves of different potato cultivars were chemically characterized by LC/MS and their antioxidant properties were investigated in vitro. Furthermore, the biological activity against the fungal pathogen Rhizoctonia solani was evaluated both in vitro and in vivo. Extracts showed the ability to inhibit R. solani growth in vitro and significantly reduced damping-off incidence in in vivo experiments. Furthermore, R. solani mycelia exposed to the extracts showed an altered morphology (low translucency, irregular silhouette, and cytoplasmatic content coagulation) compared to the untreated control in light microscopy examination. Principal component analysis conducted on identified chemical compounds highlighted significant metabolic variations across the different extracts. In particular, those that inhibited most of the growth of the pathogen were found to be enriched in α-chaconine or α-solanine content, indicating that their biological activity is affected by the abundance of these metabolites. These results clearly indicated that plant-derived compounds represent a suitable alternative to chemicals and could lead to the development of new formulates for sustainable control of plant diseases.

Bioactive compounds of different Campania native sweet pepper varieties were evaluated. Polyphenols ranged between 1.37 mmol g-1 and 3.42 mmol g-1, ß-carotene was abundant in the red variety "Cazzone" (7.05 µg g-1). Yellow and red varieties showed a content of ascorbic acid not inferior to 0.82 mg g-1, while in some green varieties the presence of ascorbic acid was almost inconsistent. Interrelationships between the parameters analyzed and the varieties showed that ascorbic acid could represent the factor mostly influencing the antioxidant activity. Polyphenol profile was different among the varieties, with a general prevalence of acidic phenols in yellow varieties and of flavonoids in red varieties. Principal Component Analysis, applied to ascorbic acid, total polyphenols and ß-carotene, revealed that two of the green varieties ("Friariello napoletano" and "Friariello Sigaretta") were well clustered and that the yellow variety "Corno di capra" showed similarity with the green varieties, in particular with "Friariello Nocerese". This was confirmed by the interrelationships applied to polyphenol composition, which let us to light on a clustering of several red and yellow varieties, and that mainly the yellow "Corno di capra" was closer to the green varieties of "Friariello".

The Black Soldier Fly (Hermetia illucens (L.), Diptera: Stratiomyidae) is an insect whose larvae thrive on agro-industrial by-products. This study reports the first use of black soldier fly larvae processing residue (BSPR) as an innovative ingredient for growing media. BSPR was characterized and evaluated to partially replace commercial peat (CP) in the production of potted plants. Chemical and microbiological analysis showed the suitability of BSPR for soilless production. Hence, six growing media mixtures (CP 100% + slow acting synthetic solid fertilizer, CP 90% + BSPR 10%, CP 80% + BSPR 20%, CP 70% + BSPR 30%, CP 60% + BSPR 40% and CP 100% without fertilizer) were assessed for the production of baby leaf lettuce, basil and tomato potted plants. Using BSPR in a proportion up to 20%, all investigated crops showed values significantly greater than or comparable to those obtained using CP 100% + slow acting synthetic solid fertilizer. In general, BSPR used in a proportion up to 20% increased the crop growth of baby leaf lettuce, basil and tomato, recording a high total dry weight (+31%, compared to the total average) and the measured leaf parameters (+39% of leaf area, +14% of leaf number), without showing abiotic stresses. This study indicates that BSPR used in a proportion up to 20% might be a valid approach for soilless production of potted baby leaf lettuce, basil and tomato plants.

Chestnut (Castanea sativa Miller.) burs (CSB) represent a solid waste produced during the edible fruit harvesting. Their usual disposal in the field increases the environmental and economic impact of the agricultural process. HPLC-UV-HRMS profiling revealed that CSB organic and aqueous extracts (CSB-M, CSB-H, CSB-A) contain several hydrolyzable tannins, mainly ellagitannins, and glycoside flavonols. Ellagic acid (EA) and chestanin are predominant components (5⁻79 and 1⁻13 mg/g dry extract, respectively). NMR analysis confirmed the chemical structures of the major constituents from CSB-M. The extracts displayed a significant scavenging activity against DPPH (EC50 12.64⁻24.94 µg/mL) and ABTS⁺ radicals (TEAC value 2.71⁻3.52 mM Trolox/mg extract). They were effective in inhibiting the mycelial growth (EC50 6.04⁻15.51 mg/mL) and spore germination (EC50 2.22⁻11.17 mg/mL) of Alternaria alternata and Fusarium solani. At the highest concentration, CSB-M was also active against Botrytis cinerea both in mycelium and spore form (EC50 64.98 and 16.33 mg/mL). The EA contributed to the antifungal activity of extracts (EC50 on spore germination 13.33⁻112.64 µg/mL). Our results can support the upgrading of chestnut burs from agricultural wastes to a resource of natural fungicides for managing fruit and vegetable diseases.

Common beans (Phaseolus vulgaris) are a good source of nutrients and phenolic compounds with versatile health benefits. Polyphenol-rich extracts of six ecotypes of P. vulgaris were analysed to determine their phenolic profiles and assayed in vitro for inhibitory effects on digestive enzymes relevant to carbohydrates and lipids metabolism. The extracts inhibited enzyme activities in a dose-dependent manner. IC 50 values ranged from 69 ± 1.9 to 126 ± 3.2 µg/mL and from 107.01 ± 4.5 to 184.20 ± 5.7 µg/mL, before and after cooking, for α-amylase, from 39.3 ± 4.4 to 74.13 ± 6.9 µg/mL and from 51 ± 7.7 to 122.1 ± 5.2 µg/mL for α-glucosidase and from 63.11 ± 7.5 to 103.2 ± 5.9 µg/mL and from 92.0 ± 6.3 to 128.5 ± 7.4 µg/mL for lipase. Results suggest encouraging their consumption, being natural sources of enzyme inhibitors important for type-2 diabetes and obesity prevention/control. Well-monitored in vivo studies would help to establish their beneficial effects, making them worthwhile of further consideration as functional foods.

Herein, we report the draft genome sequence of Pseudomonas fluorescens strain CREA-C16, a plant growth-promoting rhizobacterium that was isolated from the rhizosphere of Pisum sativum L. plants. The genome sequence is ~6 Mb in size, with a G+C content of 60.1%, and includes 4,457 candidate protein-encoding genes.

Beans are important dietary components with versatile health benefits. We analysed the extracts of twelve ecotypes of Phaseolus vulgaris in order to determine their phenolic profiles, antioxidant activity, and the in vitro antiproliferative activity. Ultra-performance liquid chromatography with diode array detector (UPLC-DAD) admitted us to detect and quantify some known polyphenols, such as gallic acid, chlorogenic acid, epicatechin, myricetin, formononetin, caffeic acid, and kaempferol. The antioxidant activity (AA) ranged from 1.568 ± 0.041 to 66.572 ± 3.197 mg necessary to inhibit the activity of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical by 50% (EC50). The extracts, except those obtained from the nonpigmented samples, were capable of inhibiting the proliferation of the human epithelial colorectal adenocarcinoma (Caco-2) cells, human breast cancer cells MCF-7, and A549 NSCLC cell line. Cultivars differed in composition and concentration of polyphenols including anthocyanins; cooking affected the antioxidant activity only marginally. Qualitative and quantitative differences in phenolic composition between the groups of beans influenced the biological activities; on the other hand, we did not find significant differences on the biological activities within the same variety, before and after cooking.

Leafy vegetables have a relatively high potential for Cd uptake and translocation, and are thus considered Cd accumulators. For this reason, leaves and roots of lettuce (Lactuca sativa L.) and endive (Cichorium endivia L.) plants, grown on different agricultural soils in Campania region (southern Italy), subjected to different fertilisation treatments (unfertilisation, compost amendment and mineral fertilisation), were analysed for Cd concentrations. Moreover, to clarify if the highest concentrations found are linked to older and inedible or to younger and edible leaves, external and internal endive leaves were separately analysed. All the leafy vegetables analysed showed on average 2-fold higher Cd concentrations in leaves than in roots. Leaf Cd concentrations in both lettuce and endive plants significantly differed among fertilisation treatments, with values highest in the plants grown on mineral fertilised soils. Apart from the soil fertilisation treatments, however, Cd leaf concentrations were often higher (up to 4-fold) than the threshold deduced by the EU 420/2011 Regulation, although the plants grew on unpolluted soils. Anyway, external leaves of endive plants showed significantly higher concentrations than internal leaves (in some cases the values were 3-fold higher), partly reassuring on the consumption of the younger leaves. Moreover, this study points out two major drawbacks in the Italian and European regulatory frameworks: (1) metal concentration (as total and/or available fraction) limits in agricultural soils are lacking; (2) metal concentration thresholds (currently existing only for Cd and Pb in crops) reported in the EU 420/2011 Regulation, expressed on the fresh weight basis rather than on the dry weight basis, appear not suitable.

The chemical composition and biological activity of the essential oils obtained from the leaves of two different cultivars of Florence fennel cropped under three different fertilization treatments (Control not fertilized; Mineral Fertilization; Compost from Municipal Solid Wastes) have been analyzed. All the oils were characterized by high anethole concentration and some showed also a good percentage of limonene. Thus, the leaves of Florence fennel, which are agricultural wastes, could be used for the recovery of anethole to be used for its flavoring and biomedical properties. The antimicrobial activity expressed by assays on the examined oils indicates an appreciable effect, generally higher on Gram-positive bacteria. The various samples of Florence fennel analyzed did not show any results with FRAP test. The DPPH test showed a weak capacity of the samples to catch the free radicals from the solution, attributable to their content in anethole.

Common bean (Phaseolus vulgaris) is the most important grain legume and plays a significant role in human nutrition being a major source of dietary protein and representing a rich source of minerals and certain vitamins. Several large germplasm collections have been established, which contain large amounts of genetic diversity, including wild and domesticated species. In this study agronomic, biochemical and molecular characterization of landrace bean named "Fagiolo occhio nero di Oliveto Citra" (Phaseolus vulgaris L.), is described. Seeds were obtained by local farmers and field trials were carried out during years 2009-2010, in the typical cultivation site (Oliveto Citra, Salerno Province), using two different densities of investment. During 2011, in order to evaluate the performance in different environments, field trials were conducted in three localities (Battipaglia, Oliveto Citra and Controne). Data analysis shows good adaptability across locations and similar grain yield using two spacing's of seeds. Morphological characterization and molecular analysis, using AFLP and Minisatellite molecular markers, were performed on ten "biotypes" collected from local farmers. Seeds characterization showed variability on the violet area surrounding the hilum (named as eye) while markers have provided useful information on relationships between biotypes. Biochemical analysis, which includes the contents of protein, minerals and antioxidants, shows how the composition is consistent with respect to other landraces and commercial cultivars. The landrace under study revealed genetic stability and good adaptation to cultivated environment with best performance in the native area. In addition, the bio-agronomic characteristics are in accord with studies reported in literature.

BACKGROUND: An ecotype of the lima bean, named 'fagiolo a Formella', which, to the best of our knowledge, is the only example of an Italian lima bean (Phaseolus lunatus L.) ecotype, is cultivated in the Campania region of southern Italy. Physical, nutritional and processing traits of dry seeds were evaluated for two consecutive growing seasons (2009 and 2010). The canning quality was also investigated, but only for the harvest of 2010. RESULTS: Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of total seed proteins allowed the attribution of 'fagiolo a Formella' to the Mesoamerican gene pool and Sieva morphotype. Seeds have a trapezoid shape, white coat and 100-seed weight greater than 42 g. Yield, protein, trypsin inhibitor and phytic acid values were found comparable with those reported for lima bean varieties cultivated in sub-tropical areas. Moreover, we found that this ecotype is devoid of lectin. CONCLUSIONS: The good adaptation to growing environment is indicated by the fact that 'fagiolo a Formella' seed quality is comparable to that of lima beans grown in America. Overall the canning quality was found satisfactory and canning significantly destroys the main anti-nutritional compounds present in dry seeds.

Recently, genome sequencing of many isolates of genetically monomorphic bacterial human pathogens has given new insights into pathogen microevolution and phylogeography. Here, we report a genome-based micro-evolutionary study of a bacterial plant pathogen, Pseudomonas syringae pv. tomato. Only 267 mutations were identified between five sequenced isolates in 3,543,009 nt of analyzed genome sequence, which suggests a recent evolutionary origin of this pathogen. Further analysis with genome-derived markers of 89 world-wide isolates showed that several genotypes exist in North America and in Europe indicating frequent pathogen movement between these world regions. Genome-derived markers and molecular analyses of key pathogen loci important for virulence and motility both suggest ongoing adaptation to the tomato host. A mutational hotspot was found in the type III-secreted effector gene hopM1. These mutations abolish the cell death triggering activity of the full-length protein indicating strong selection for loss of function of this effector, which was previously considered a virulence factor. Two non-synonymous mutations in the flagellin-encoding gene fliC allowed identifying a new microbe associated molecular pattern (MAMP) in a region distinct from the known MAMP flg22. Interestingly, the ancestral allele of this MAMP induces a stronger tomato immune response than the derived alleles. The ancestral allele has largely disappeared from today's Pto populations suggesting that flagellin-triggered immunity limits pathogen fitness even in highly virulent pathogens. An additional non-synonymous mutation was identified in flg22 in South American isolates. Therefore, MAMPs are more variable than expected differing even between otherwise almost identical isolates of the same pathogen strain.

BACKGROUND: Grass pea seeds are a good source of vegetable proteins, but the presence of toxic and antinutritional compounds represents a barrier to their large-scale use as food or animal feed. How much growing location and/or seasonal climate might affect the storage of these factors has been little investigated. RESULTS: Fourteen Italian ecotypes of grass pea were cultivated in two locations in southern Italy characterised by different climatic conditions. The seven ecotypes with the best yields and/or seed quality were investigated for a further two growing seasons. From a statistical point of view the physicochemical and nutritional traits among ecotypes were not the same from one year to the next. Moreover, a significant positive correlation was found between ß-oxalyl-diamino-propionic acid and trypsin inhibitor contents. The lowest levels of both these compounds were associated with the highest amount of rainfall during the plant vegetative cycle. CONCLUSION: Principal component analysis of the data showed that the overall seed composition was affected by the growing location. Consequently, each grass pea genotype should also be carefully investigated in relation to different environments before being considered for release as safe for widespread human or animal consumption.

The temporal dynamics of some trace elements in two different types of Mediterranean soils were studied in order to evaluate the possible long-term contamination following compost amendments. Total and available (DTPA-extractable) concentrations of Cd, Cu, Pb and Zn were determined. The study was carried out on two agricultural soils in Campania region (southern Italy), a Sandy Loam Calcaric Cambisol (SG) and a Clay Gleyc Luvisol (CO), during 3 years of organic amendment with compost. The compost, produced from the organic fraction of municipal solid waste and urban yard trimmings, in accordance with the Italian law for agricultural use, was applied at annually rates of 15, 30, and 45 t ha(-1) (on dry weight basis). Wide variations in total and available Cd, Cu, Pb and Zn concentrations were observed over time, but appeared to be in many cases unrelated to the treatments, occurring also in control plots. After 3 years of compost application the amended SG soil showed the highest and significant increase in total Cd and Zn concentrations; in addition, the available Cd, Pb and Zn concentrations increased in accordance with the compost rates. The CO soil, characterized by a higher clay content, lower organic matter content and lower cation exchange capacity, exhibited a lower increase in available metal fractions. Our findings show that compost amendment affects more the available than the total metal concentrations in the two types of soils studied and thus it is important into legislation that metal "bioavailability" may be considered in defining threshold metal values.
